<details>Micrographs of CVA9 treated with endosomal ionic condition buffer for 3 hours at 37 °C. Sample was applied to Quantifoil R1.2/1.3 holey carbon grids with 300 copper mesh and manually blotted and flash-frozen using a homemade plunger. Data was acquired at the Biocenter Finland National Cryo-EM facility with a FEI TALOS Artica transmission electron microscope operated at 200 kV. Movies were collected with a Falcon III direct electron detector at a nominal magnification of 120 000x giving a pixel size of 1.24 Å per pixel. Each movie consisted of 30 frames with an exposure time of 1 second per frame. The total electron dose was approximately 30 electrons per Å2. Movies were corrected for beam-induced motion by aligning and averaging all frames with Motioncor2 implemented in Scipion version 2.0.</details>
